Abstract
We studied the therapeutic effect of 5-fluoruracil in combination with low dose leucovorin in patients with recurrent or metastatic carcinoma of the colon or rectum. 40 patients with clearly measurable lesions were entered into this study between

1989 to May 1991 and two were excluded from the analysis because one patient did not meet the eligibility criteria and the other was lost to follow-up. They were treated with leucovorin, 20mg/m*/day iv bolus, followed by 5-fluorouracil

bolus for 5 consecutive days. This regimen was repeated every 4 weeks and 5-fluourouracil doses were escalated to 420mg/m*/day if the previous dose was tolerable. Male to female ratio was 25 to 13 with median age of 54 years(32~81 years).22
patients had
previous history of chemotherapy and 16 were previously untreated.
There was no complete and the partial response rate was 26.4%(10 of 38 patients).
The median response duration was 5 months and the median tme to progression was 3 months. Hematologic toxicities were mild and the rates of grade 3 or greater nonhematologic toxicity were 7.8%(3 of 38 patients). There was no treatment-related
fatality.
Our results indicate that the combination of 5-fluorouracil and leucovorin has modest activity for patients with metastatic or recurrent carcinoma of the rectum and colon.
